Stress Tolerance Effect of Dora AOS on Lettuce

In the ever-evolving landscape of agriculture, the quest for effective solutions to enhance crop resilience against various stresses is of paramount importance. A comprehensive study was conducted at the National Agriculture Park Changshu (CSNAP) to evaluate the efficacy of Dora AOS powder in improving lettuce’s tolerance to drought, waterlogging, and salinization stresses. The experiment involved six treatment groups, with five lettuce plants in each group. ‘Italy’ lettuce was used as the test material, and the AOS powder was applied at a concentration of 50ppm in the treatment groups, while the control groups were treated with water under different stress conditions.

Experimental Setup

Lettuce seeds were carefully sterilized with 75% (v/v) alcohol for 15 minutes and then rinsed thoroughly with distilled water. They were sown in seedling trays, and after transplanting, the plants were watered with either clear water or a 50ppm ArouseTM solution on the 1st and 10th day. Stress simulation treatments commenced 14 days after transplanting:

Drought Stress: The soil moisture level was maintained at a low level from day 14 to day 22 after transplanting.

Waterlogging Stress: On the 14th day of transplanting, a 2cm waterlogging layer was created and maintained for 4 days. After applying the treatment solutions, the waterlogging state continued for another 4 days.

Salinization Stress: From the 14th day of transplanting, each pot received 150 mL of a 4% (w/v) NaCl solution through root irrigation, and the salt stress state was maintained for 4 days, followed by another 4 days of the same salinity level.

How To Measure

After the treatment period, the plants were harvested, and key growth parameters were measured. These included plant height, stem diameter, plant weight, and relative water content. The relative water content was calculated using a specific formula:
(X1−X2)/(X3−X2), where X1 is the fresh mass excluding the roots, X2 is the mass after curing at 105 °C for 15 minutes and drying at 80 °C, and X3 is the mass when saturated with water after immersion in distilled water at 4 °C for 24 hours. Photographs were also taken to document the growth process and compare the performance of each group.

Impressive Results

The data analysis revealed significant differences between the control and treatment groups, highlighting the positive impact of Dora AOS on lettuce growth under stress conditions.

Drought Stress: The AOS-treated seedlings in group exhibited a 11.85% increase in plant height compared to the control group . The plant weight was also significantly higher, with an additional 13.1 g. The relative water content of the treated plants was 2.47% higher, indicating enhanced osmotic regulation and drought resistance.

Waterlogging Stress: Although the plant height remained stable, the lettuce plants in the treatment group had a 4.52 g higher weight compared to the control group. The relative water content was 3.87% higher, suggesting improved waterlogging tolerance.

Salinization Stress: In the salinization stress treatment, the AOS treated group showed a higher stem diameter compared to the control group. The plant weight was significantly higher, with an average weight of 34.0 g, indicating a clear growth advantage. The relative water content was also substantially higher, reaching 83.04%, compared to the control group’s 69.23%.

Conclusion

The results of this trial clearly demonstrate that Dora AOS is highly effective in enhancing lettuce’s tolerance to drought, waterlogging, and salinization stresses. It shows excellent performance in promoting plant height, increasing plant weight, and maintaining a high relative water content. Based on these findings, it is recommended to apply Dora AOS at a dosage of 50ppm to boost plants’ abilities to resist abiotic stresses.
